Bird Flu Mutation Study Offers Vaccine Clue

Sciencedaily — Scientists have described small genetic changes that enable the H5N1 bird flu virus to replicate more easily in the noses of mammals.

So far there have only been isolated cases of bird flu in humans, and no widespread transmission as the H5N1 virus can't replicate efficiently in the nose. The new study, using weakened viruses in the lab, supports the conclusions of controversial research published in 2012 which demonstrated that just a few genetic mutations could enable bird flu to spread between ferrets, which are used to model flu infection in humans.
Researchers say the new findings could help to develop more effective vaccines against new strains of bird flu that can spread between humans.
"Knowing why bird flu struggles to replicate in the nose and understanding the genetic mutations that would enable it to happen are vital for monitoring viruses circulating in birds and preparing for an outbreak in humans," said Professor Wendy Barclay, from the Department of Medicine at Imperial College London, who led the study.
"The studies published last year pointed to a mechanism that restricts replication of H5N1 viruses in the nose. We've engineered a different mutation with the same effect into one of the virus proteins and achieved a similar outcome. This suggests that there is a common mechanism by which bird flu could evolve to spread between humans, but that a number of different specific mutations might mediate that."
Bird flu only rarely infects humans because the human nose has different receptors to those of birds and is also more acidic. The Imperial team studied mutations in the gene for haemagglutinin, a protein on the surface of the virus that enables it to get into host cells. They carried out their experiments in a laboratory strain of flu with the same proteins on its surface as bird flu, but engineered so that it cannot cause serious illness.
The research found that mutations in the H5 haemagglutinin enabled the protein to tolerate higher levels of acidity. Viruses with these mutations and others that enabled them to bind to different receptors were able to replicate more efficiently in ferrets and spread from one animal to another.
The results have important implications for designing vaccines against potential pandemic strains of bird flu. Live attenuated flu vaccines (LAIV) might be used in a pandemic situation because it is possible to manufacture many more doses of this type of vaccine than of the killed virus vaccines used to protect against seasonal flu. LAIV are based on weakened viruses that don't cause illness, but they still have to replicate in order to elicit a strong immune response. Viruses with modified haemagglutinin proteins induced strong antibody responses in ferrets in this study, suggesting that vaccines with similar modifications might prove more effective than those tested previously.
"We can't predict how bird flu viruses will evolve in the wild, but the more we understand about the kinds of mutations that will enable them to transmit between humans, the better we can prepare for a possible pandemic," said Professor Barclay.
The research was funded by the Medical Research Council and the Wellcome Trust and published in the Journal of General Virology.

'Tôi chắc chắn công an đánh tôi'

Tiến sỹ Nguyễn Quang A

WikiLeaks Makes Leaked US Documents Searchable

Meredith Buel
ulian Assange speaks during a teleconference between London and Washington on April 8, 2013
The whistleblowing website WikiLeaks has launched a new database containing 1.7 million documents from the U.S. State Department that were declassified, but were difficult for the public to access.
WikiLeaks is calling the searchable collection the “Public Library of U.S. Diplomacy," bringing together diplomatic and intelligence documents that previously could only be accessed through the National Archives.
WikiLeaks founder Julian Assange told reporters via video link from the Ecuadorian embassy in London that the documents were hidden in what he called the borderline between secrecy and complexity.
“This material that we have published today is the single most significant geopolitical publication that has ever existed," he said.

The database gives the public access to diplomatic cables from the beginning of 1973 to the end of 1976, including communications sent by then-U.S. Secretary of State Henry Kissinger.

At a media briefing in Washington, WikiLeaks spokesman Kristinn Hrafnsson called the new database a public service.

“In my mind when you look at this material, 1.7 million public documents, they are not easily available.  It is extremely hard to approach them even though they have been declassified.  So making them available to people is basically taking the secrecy away and uncovering the stories," she said.

Although the documents have long been in the public domain, their release in a searchable archive has generated some headlines internationally because their publication was coordinated with a number of media outlets.

For example, India’s Hindu newspaper cited the cables in a report about Rajiv Gandhi, whose family still dominates the country’s ruling party, as a middleman for an arms deal in the 1970s.

Gandhi was assassinated in 1991and his wife Sonia is now head of the ruling Congress party.

U.S. State Department spokesman Patrick Ventrell declined comment on the archive. “Our understanding is that there are some very old documents here.  We are still looking to see what all of this may be…I cannot comment on neither their authenticity nor their status of classification," he said.

The new WikiLeaks database contains 250,000 classified cables leaked by the website in 2010.

Those documents infuriated the international community as they provided blunt and unflattering U.S. views of world leaders.

Assange has been seeking refuge in Ecuador’s embassy in London in an effort to avoid extradition to Sweden to face allegations of rape and sexual assault, accusations Assange has denied.

U.S. Army Private Bradley Manning admitted to giving WikiLeaks the first set of cables.  His court martial is set to begin in June.

New Brain-Test App

Smartphones offer a wealth of possibilities for psychological research. A new study shows that an iPhone app yields as reliable results as laboratory tests.

Two years ago, researcher Josef Bless was listening to music on his phone when he suddenly had an idea. "I noticed that the sounds of the different instruments were distributed differently between the ears, and it struck me that this was very similar to the tests we routinely use in our laboratory to measure brain function. In dichotic listening, each ear is presented with a different syllable at the same time (one to the left and one to the right ear) and the listener has to say which syllable seems clearest. The test indicates which side of the brain is most active during language processing," Bless explains.
Josef Bless is working on a PhD in psychology at the University of Bergen, Norway. He is a member of the Bergen fMRI Group, an interdisciplinary research group headed by Professor Kenneth Hugdahl, who has received a European Research Council (ERC) Advanced Grant for his brain research.
The iPhone app for dichotic listening is called iDichotic and was launched on the App Store in 2011, where it can be downloaded for free. Some one year later, more than 1,000 people have downloaded the app, and roughly half have sent their test results to the researchers' database.
The researchers analysed the first 167 results they received and compared them with the results of 76 individuals tested in laboratories in Norway and Australia. The results have been published in the journal Frontiers in Psychology.
"We found that the results from the app were as reliable as those of the controlled laboratory tests. This means that smartphones can be used as a tool for psychological testing, opening up a wealth of exciting new possibilities," says Bless.
"The app makes it possible to gather large volumes of data easily and inexpensively. I think we will see more and more psychological tests coming to smartphones," he adds.
The researchers have also developed a special version of iDichotic for patients with schizophrenia who suffer from auditory hallucinations (i.e. hear "voices"). The app helps in training patients to improve their focus, so that when they hear voices, they are better able to shut them out.
"Using a mobile app, patients can be tested and receive training at home, instead of having to come to our laboratory," says Bless.
The app iDichotic has been developed in collaboration with Professor Kenneth Hugdahl, Doctor René Westerhausen, and Magne Gudmundsen.
Test yourself You can download iDichotic from the App Store. The listening test takes three minutes and tells you which side of your brain is most active in language processing. Most people primarily use the left side of the brain, but for a minority (including many left-handed people) the right side of the brain is more involved in language processing. In addition, the test measures attention when the task is to focus on one ear at a time. You can send your results to the researchers if you want. iDichotic is currently available for iOS devices.

Computer Scientists Develop Video Game That Teaches How to Program in Java

Apr. 8, 2013 — Computer scientists at the University of California, San Diego, have developed an immersive, first-person player video game designed to teach students in elementary to high school how to program in Java, one of the most common programming languages in use today.
The researchers tested the game on a group of 40 girls, ages 10 to 12, who had never been exposed to programming before. They detailed their findings in a paper they presented at the SIGCSE conference in March in Denver. Computer scientists found that within just one hour of play, the girls had mastered some of Java's basic components and were able to use the language to create new ways of playing with the game.
"CodeSpells is the only video game that completely immerses programming into the game play," said William Griswold, a computer scientist at the Jacobs School of Engineering at UC San Diego.
The UC San Diego computer scientists plan to release the game for free and make it available to any educational institution that requests it. Researchers are currently conducting further case studies in San Diego elementary schools.
Teaching computer science below the college level is difficult, mainly because it is hard to find qualified instructors for students in elementary to high school, Griswold said. So he and his graduate students set out to find a way to reach these students outside the classroom. They designed the game to keep children engaged while they are coping with the difficulties of programming, which could otherwise be frustrating and discouraging.
Teaching children how to program must be a priority in a society where technology is becoming more and more important, said Sarah Esper, one of the lead graduate students on the development of CodeSpells. Programming also teaches logical thinking, said Stephen Foster, another lead student.
"We're hoping that they will get as addicted to learning programming as they get addicted to video games," Foster said.
How CodeSpells works
CodeSpells' story line is simple: the player is a wizard arriving in a land populated by gnomes. The gnomes used to have magic, but lost it at some point. The wizard must help them. She (or he) writes spells in Java. Players have seven spells available to them, including levitating objects within the game, flying and making fire.
Players can also earn badges by undertaking simple quests, which help them master the game's spells. One quest entails crossing a river. Another entails rescuing a gnome from the roof of his cottage, where he got stuck. Yet another entails starting a large bonfire. By the time players complete the game's first level, they have learned the main components of the Java programming language, such as parameters, for if statements, for loops and while loops, among other skills.
Testing the game
Researchers tested the game on a group of 40 girls ages 10 to 12 in San Diego. They gave the students a brief overview of the game's mechanics, including how to write and edit code within the game's user interface. The girls were divided in groups of two or three. Researchers encouraged them to explore the game and see what they could do. "We were purposefully vague," they wrote, "as we hoped to encourage a largely unstructured learning environment."
The students were disappointed when they had to stop playing because the test was over. Their interest in the game didn't wane when they made mistakes while writing code. Instead, they used the mistakes as a stepping stone to explore the game's possibilities. For example, one group made the mistake of levitating an object so high into the air that their wizard couldn't reach it. So the girls made their wizard jump onto another object and levitated it high enough to reach the object they were after. The girls also reported feeling empowered. When they encountered a difficulty, they tried different spells and made changes to the code until they solved it.
Computer science learning theory
CodeSpells was influenced by research that Esper and Foster conducted on how successful programmers learn their trade. They surveyed 30 computer scientists and identified five characteristics that are key to learn programming outside a classroom setting: activities must be structured by the person who is trying to learn; learning must be creative and exploratory; programming is empowering; learners have difficulty stopping once they start; and learners spend countless hours on the activity.
Researchers summarized these findings in their SIGCSE 2013 paper, humorously titled "On the Nature of Fires and How to Spark Them When You're Not There."
Esper will present her CodeSpells work April 18 at Research Expo at the Jacobs School of Engineering at UC San Diego.

China’s attack on fishing boat is inhuman: law experts

he captain of the fishing boat that was fired at by a Chinese naval ship, Pham Quang Thanh, is seen on board the fired boat

Many Vietnamese law experts have condemned China’s recent pursuit and subsequent firing at a Vietnamese fishing boat in the waters off Vietnam’s Hoang Sa (Paracel) archipelago. They consider the incident to be inhuman and boldly in violation of Vietnam’s sovereignty.
Commenting on the incident, which involved a fishing boat from the central province of Quang Ngai on March 20, lawyer Truong Xuan Tam, a member of the National Lawyers Council and deputy president of the Ba Ria-Vung Tau Province Bar Association, pointed out that China has not stopped at threatening to use force, but it has actually used force in violating Vietnam’s sovereignty.
“We need to consider taking proceedings against China to the International Court of Justice to claim compensation for Vietnamese fishermen for their damage and demand that China respect Vietnam’s sovereignty,” the lawyer said.
Tam condemned the incident as extremely dangerous and a bold, rough and defiant violation of Vietnam’s sovereignty.
Another lawyer, Hoang Ngoc Giao, head of the Research Institute for Policy, Law and Development, said: “No armed conflict has taken place between China and Vietnam, but China has used force towards Vietnamese civilians. As far as I know, no country has behaved as badly as China in this case.”
The former Vietnamese consul general to China’s Guangzhou Province, Duong Danh Hy, also condemned the aggressive action. “The attack on fishermen is a bold escalation by China in violating Vietnam’s sovereignty. Besides the objections made through diplomatic channels, we must pay more attention to protecting the lives of our fishermen.”
Nguyen Ngoc Duc, Chief of the Secretariat of the Vietnamese Fishery Association, said the organization has proposed that relevant Vietnamese agencies take measures to “prevent wrong actions by China, safeguard the life and property of Vietnamese fishermen, and protect Vietnam’s sovereignty over its seas and islands.”
As previously reported, the QNg 96382 TS fishing boat from Quang Ngai was conducting normal fishing activity in the traditional fishing ground around the Hoang Sa archipelago when it was chased and shot at by Chinese vessels on March 20. The assault caused a fire that burned the boat’s cabin.
In answering reporters’ queries regarding Vietnam’s response to the incident, Foreign Ministry Spokesman Luong Thanh Nghi said this was an extremely serious act that violated Vietnam’s sovereignty over the archipelago and threatened the lives of, and caused damage to, Vietnamese fishermen.

It also seriously violates the spirit of the Declaration on the Conduct of Parties in the East Sea (DOC), breaches the principles of international law and the agreement on the basic principles guiding the settlement of sea issues between Vietnam and China, Nghi added.
Meanwhile, at a press conference on March 26, Chinese foreign ministry spokesman Hong Lei said that China had taken unspecified but "legitimate and reasonable" actions against Vietnamese boats working “illegally in Chinese waters”. He denied that any boats had been damaged.
In a statement released the same day, the Chinese Defense Ministry also refuted the incident and said that a Chinese navy vessel had fired two flares into the air after four Vietnamese fishing boats failing to respond to whistles, shouts and signal flags requesting that they stop fishing and leave the area, which China claims as its territorial waters.
